Methods are provided for purification of Factor VIII polypeptides by immunoaffinity chromatography and ion exchange chromatography, in which the eluate from the immunoaffinity column is diluted with a solution comprising higher salt concentration, or lower non-polar agent concentration than that of the elution solution, prior to passing the diluted solution through the ion exchange column. The methods result in improved purification without significant yield loss.
L'invention concerne des méthodes permettant de purifier des polypeptides du facteur VIII par chromatographie d'immunoaffinité et chromatographie d'échange d'ions. Selon ces méthodes, l'éluat provenant de la colonne d'immunoaffinité est tout d'abord dilué à l'aide d'une solution, qui présente une concentration en sel élevée ou une concentration d'un agent non polaire inférieure à celle de la solution d'élution. Ces méthodes, qui consistent ensuite à faire passer la solution diluée à travers la colonne d'échange d'ions, permettent d'obtenir une purification améliorée sans baisse de rendement importante.
