Improved processes for the synthesis of 2'-O-substituted pyrimidine nucleosides and oligomeric compounds having these nucleosides are disclosed. The synthetic processes feature alkylation of a 2,2'-anhydropyrimidine nucleoside or a 2S,2'-anhydropyrimidine nucleoside with a weak nucleophile in the presence of a Lewis acid.
L'invention concerne un procédé amélioré pour la synthèse des nucléosides pyrimidines à substitution 2'-O et des composés oligomères renfermant ces nucléosides. Le procédé de synthèse repose sur l'alkylation d'une 2,2'-anhydropyrimidine nucléoside ou d'une 2S,2'-anhydropyrimidine nucléoside, avec utilisation d'un nucléophile faible en présence d'acide de Lewis.
