Improved results are obtained using in an enzyme fragment complementation assay, as an enzyme donor a fragment of .beta.-galactosidase of from 36 to 50 amino acids joined to a ligand capable of binding to a receptor other than an immunoglobulin or fragment thereof. Conveniently, for enzyme assays, enzyme binding site inhibitors are conjugated to the enzyme donor, whereby binding of the enzyme to the binding site inhibitor results in a reduction in the turnover rate of .beta.-galactosidase in the presence of EA and a substrate capable of producing a detectable signal. Analogously, ligands for membrane receptors may be conjugated to the ED for measurement of membrane receptors.
