An improved double-stranded RNA molecule having been improved to thereby control the gene expression inhibitory effect of an siRNA. Namely, a double- stranded RNA molecule capable of inhibiting the expression of a target gene in a cell by RNAi wherein one or more nucleotides from the 3~- or 5~-end of the sense strand in the double-stranded part are converted into nucleotides which are not complementary to the antisense strand. In the sense strand of the double-stranded part of this double-stranded RNA molecule, moreover, the number of the nucleotides which are complementary to the antisense strand enables the hybridization of both strands in the above-described cell.
La présente invention concerne une molécule d'ARN double brin ayant été améliorée de façon à commander ainsi un effet inhibiteur de l'expression de gène d'un ARNsi. A savoir, une molécule d'ARN double brin capable d'inhiber l'expression d'un gène cible dans une cellule par un ARNi, dans lequel une ou plusieurs nucléotides de l'extrémité 3' ou 5' du brin sens dans la partie double brin sont transformés en nucléotides qui ne sont pas complémentaires du brin antisens. Par ailleurs, dans le brin sens de la partie double brin de cette molécule d'ARN double brin, le nombre de nucléotides complémentaires du brin antisens permet l'hybridation des deux brins de la cellule susmentionnée.
