Disclosed is a recombinant viral particle capable of delivering a nucleic acid to mammalian cells, said particle comprising a member of a first specific binding pair capable of binding to a first receptor expressed on the surface of a cell so as to cause infection thereof, and a surface-exposed member of a second specific binding pair capable of binding to a second receptor generally not expressed on the surface of the cell, such that binding of the viral particle to the second receptor via the member of the second specific binding pair inhibits infection of the cell by the viral particle. Also disclosed is a composition comprising the recombinant viral particle defined above, and methods of altering the host range of a viral particle.
L'invention se rapporte à une particule virale recombinante permettant d'apporter un acide nucléique à des cellules de mammifère, et comprenant un élément d'une première paire de liaison spécifique apte à se lier à un premier récepteur exprimé à la surface d'une cellule, de manière à infecter cette dernière, et un élément, exposé à la surface de la particule, d'une seconde paire de liaison spécifique apte à se lier à un second récepteur qui n'est généralement pas exprimé sur la surface de la cellule, de sorte que la liaison de la particule virale au second récepteur par l'élément de la seconde paire de liaison spécifique inhibe l'infection de la cellule par la particule virale. L'invention se rapporte également à une composition comprenant la particule virale recombinante définie ci-dessus, ainsi qu'à des procédés de modification de la gamme d'hôtes d'une particule virale.
