A method for in situ dissolution of an alginate coating from transplants containing biological tissue cores by administering to a transplant's recipient a physiologically acceptable chelating agent in an amount sufficient to dissolve the alginate coating is disclosed. The method allows in situ removal of the coating and deactivation of the transplant by rejection of its core by the host immune system without the necessity of having to perform surgery. The figure illustrates the blood glucose values observed before and after a cluster method dissolution of single alginate coating of rat pancreatic islets implanted in the peritoneal cavity of streptozotocin-induced diabetic mice to treat induced diabetes.
L'invention a pour objet un procédé de dissolution in situ de couches d'alginate provenant de greffes contenant des noyaux de tissus biologiques. Ledit procédé consiste à administrer à un sujet greffé un agent chélateur, physiologiquement acceptable, dans une quantité suffisante pour dissoudre la couche d'alginate. Ce procédé permet de retirer la couche in situ et de désactiver la greffe en provoquant le rejet de son noyau par le système immunitaire hôte sans nécessiter le recours à la chirurgie. La figure ci-dessous présente les taux de glycémie observés avant et après dissolution, selon un procédé en grappes, d'une seule couche d'alginate d'îlots pancréatiques de rat implantés dans la cavité péritonéale d'une souris diabétique par induction de streptozotocine, afin de traiter le diabète induit.
