The invention describes a method for isolating one or more genetic elements encoding a gene product having a desired activity, comprising the steps of: (a) compartmentalising genetic elements into microcapsules; and (b) sorting the genetic elements which express the gene product having the desired activity; wherein at least one step is under microfluidic control. The invention enables the in vitro evolution of nucleic acids and proteins by repeated mutagenesis and iterative applications of the method of the invention.
La présente invention concerne un procédé permettant d'isoler un ou plusieurs éléments génétiques codant un produit génique doté d'une activité attendue. Pour ce procédé, on commence (a) par compartimenter les éléments génétiques en microcapsules, puis (b) on trie les éléments exprimant le produit génique doté de l'activité attendue, l'une au moins des deux opérations se déroulant sous commande microfluide. L'invention permet ainsi de faire évoluer in vitro des acides nucléiques et des protéines par mutagenèse répétée et applications itératives du procédé de l'invention.
