Compositions and methods are provided for the enhanced in vitro synthesis of polypeptides. In order to improve the performance of in vitro protein synthesis reactions, metabolic inhibitors, or manipulation of a source organism, is used to diminish or avoid the action of enzymes responsible for undesirable amino acids production or depletion. A homeostatic system may be used for production of ATP, where the required high energy phosphate bonds are generated in situ, e.g. through coupling with an oxidation reaction. The homeostatic energy source will typically lack high energy phosphate bonds itself, and will therefore utilize free phosphate in the reaction mix during generation of ATP. The homeostatic energy source is provided in combination with an enzyme that catalyzes the creation of high energy phosphate bonds and with an enzyme that can use that high energy phosphate bond to regenerate ATP.

L'invention concerne des compositions et des techniques destinées à la synthèse in vitro améliorée des polypeptides. Afin d'améliorer les performances des réactions de synthèse in vitro de protéines, on utilise des antimétabolites, ou une manipulation d'un organisme source, en vue de diminuer ou d'éviter l'action des enzymes responsables de la production ou de la déplétion non recherchées d'acides aminées. On peut utiliser un système homéostatique pour la production d'ATP, les liaisons phosphate à haute énergie nécessaires étant générées in situ, notamment par le couplage avec une réaction d'oxydation. La source d'énergie homéostatique ne contient généralement pas ces liaisons phosphate à haute énergie et elle utilise par conséquent des phosphates libres dans le mélange réactionnel au cours de la production d'ATP. La source d'énergie homéostatique est fournie en combinaison avec une enzyme catalysant la création de liaisons phosphate à haute énergie, et une enzyme pouvant utiliser lesdites liaisons phosphate aux fins de produire de l'ATP.
