It is intended to provide a screening system for a drug which passes the blood- brain barrier and acts on the center, a drug which acts on the blood-brain barrier per se, or a drug which is not expected to act in the center but migrates into the brain. Further, another object of the invention is to realize a pathogenesis analysis research under pathologic conditions or the screening by imposing various pathologic environments on the screening system. A blood-brain barrier in vitro model is prepared by using a three-dimensional culture device comprising a culture medium, a plate for holding the culture medium and a filter soaked in the culture medium and placed such that it does not come into contact with the bottom surface of the plate, wherein the filter has many pores with a diameter of from 0.35 to 0.45 µm, and inoculating a primary cultured brain capillary endothelial cell on the upper surface of the filter, inoculating a primary cultured brain pericyte on the lower surface of the filter, inoculating a primary cultured astrocyte on the inner surface of the plate and co-culturing them with a standard culture medium.
La présente invention concerne un système de criblage destiné à un médicament qui traverse la barrière hémato-encéphalique et qui agit sur la partie centrale, un médicament qui agit sur la barrière hémato-encéphalique per se ou un médicament qui n~est pas supposé agir sur la partie centrale mais qui migre vers le cerveau. En outre, l~invention se rapporte à une analyse de la pathogénie dans des conditions pathologiques ou au criblage en imposant au système de criblage différents environnements pathologiques. Un modèle in vitro de barrière hémato-encéphalique est préparé en : utilisant un dispositif de culture tridimensionnel pourvu d~un milieu de culture, d~une plaque servant à contenir le milieu de culture ainsi que d~un filtre introduit dans le milieu de culture et disposé de manière à ne pas venir au contact de la surface inférieure de la plaque, le filtre possédant de nombreux pores dont le diamètre est compris entre 0,35 et 0,45 µm ; et en inoculant une cellule endothéliale de capillaire cérébral de culture primaire sur la surface supérieure du filtre, un péricyte cérébral de culture primaire sur la surface inférieure du filtre, un astrocyte de culture primaire sur la surface interne de la plaque et en les co-cultivant dans un milieu de culture standard.
