Compounds comprising two human GlxI inhibitors covalently linked via a chemical linker are provided, wherein each of said two human GlxI inhibitors, which may be the same or different, is an S-substituted glutathione or an S- substituted glutathione prodrug, wherein said GlxI inhibitors each have a .gamma.-glutamyl amino group, wherein said chemical linker is covalently bound to each GlxI inhibitor via said .gamma.-glutamyl amino group, and wherein said chemical linker has a length of at least 50 Angstroms. Monovalent irreversible inactivators of human GlxI are also provided. An antineoplastic composition is provided, which comprises a compound described above and a pharmaceutically acceptable carrier.In vitro and in vivo methods of preventing or inhibiting the growth and proliferation of neoplastic cells and/or tumors are also provided.

L'invention concerne des composés comprenant deux inhibiteurs GlxI humains reliés de manière covalente par un liant chimique. Chacun de ces inhibiteurs peut être identique ou différent par rapport à l'autre, et est un glutathione S-substitué ou un promédicament de glutathione S-substitué. Ces inhibiteurs présentent chacun un groupe amino .gamma.-glutamyle. Chaque liant chimique est relié de manière covalente à chaque inhibiteur GlxI, par ledit groupe amino .gamma.-glutamyle, et chaque liant chimique présente une longueur d'au moins 50 .ANG.nsgtroms. Des inactivateurs irréversibles monovalents de GlxI humains sont également décrits. L'invention concerne une composition antinéoplastique comprenant un composé décrit ci-dessus et un vecteur pharmaceutiquement acceptable. L'invention concerne des méthodes <i>in vitro </i>et<i> in vivo</i> de prévention ou d'inhibition de la croissance et de la prolifération de cellules néoplastiques et/ou de tumeurs.
