Cell lines and methods are disclosed for detect- ing the presence of RNA viruses in a specimen. The cell lines are stably transformed with a DNA mole- cule that includes a promoter capable of being recog- nized by the DNA dependent RNA polymerase of the cell capable of directing the transcription of a cDNA of a structurally defective RNA virus genome oper- ably coupled to the promoter. The cDNA contains a structural coding sequence encoding a selected re- porter gene product. The RNA molecules transcribed by the DNA dependent RNA polymerase are not ca- pable of causing the translation of the reporter gene in the cell except when an active related virus that provides the necessary trims-acting enzymes to cause the increased replication of the RNA containing the reporter gene which is then translated into the re- porter gene product is provided. Methods utilizing the cell lines of this invention to detect RNA viruses in a specimen by incubating the specimen with the cell line and assaying for expression of the reporter gene and a kit containing a supply of the cells and a supply of the reagents necessary for the detection of the reporter gene product are also provided.
L'invention a pour objet des lignées cellulaires et des procédés destinés à détecter la présence de virus à ARN dans un spécimen. Les lignées cellulaires sont transformées de façon stable avec une molécule d'ADN comprenant un promoteur pouvant être reconnu par l'ARN-polymérase ADN-dépendante de la cellule pouvant diriger la transcription d'un ADNc d'un génome de virus à ARN structurellement défectif, couplé de façon fonctionnelle au promoteur. L'ADNc renferme une séquence structurelle de codage d'un produit génique indicateur sélectionné. Les molécules d'ARN transcrites par l'ARN-polymérase ADN-dépendante ne peuvent pas entraîner la traduction du gène indicateur dans la cellule, sauf si l'on dispose d'un virus actif fournissant les enzymes transactives nécessaires pour accroître la réplication de l'ARN contenant le gène indicateur transcrit ensuite dans le produit génique indicateur. L'invention décrit également des procédés utilisant lesdites lignées cellulaires afin de détecter les virus à ARN dans un spécimen par incubation du spécimen avec la lignée cellulaire et par des méthodes d'expression du gène indicateur et d'un ensemble contenant un apport de cellules et un apport d'agents réactifs nécessaires à la détection du produit génique indicateur.
