The present invention provides an indolomorphinan derivative or a pharmacologically acceptable acid addition salt thereof, and an agent for curing and preventing cerebral disorders composed of the derivative and salt thereof, which is represented by the following formula (I): (see formula I) [wherein R1 represents cyclopropylmethyl or the like; R2 and R3 each represent hydroxy, methoxy, or the like; R4 represents hydrogen, methyl, benzyl, 3-isothiocyanatopropyl, or the like; and (R5)m represents hydrogen, substituted benzo, or the like]. It was apparent that the compounds of the present invention have the excellent effect of preventing damages of the cerebral nerve cells. Therefore, the compounds of the present invention are useful as medicines used for curing and preventing various cerebral diseases such as cerebral stroke, traumatic cerebral diseases, cerebral edema, and cerebral neurodegenerative diseases, ameliorating aftereffects thereof, and preventing the recurrence thereof by inhibiting various ischemic, hemorrhagic or traumatic cerebral disorders, and damages of the cerebral nerve cells caused by various nerve degenerations.
Cette invention se rapporte à des dérivés d'indolomorphinane, représentés par la formule générale (I) ou à des sels d'addition d'acide de ces composés, acceptables sur le plan pharmacologique, et à des médicaments/remèdes préventifs contre les troubles cérébraux qui contiennent ces composés. Dans ladite formule, R<1> représente cyclopropylméthyle, etc.; R<2> et R<3> représentent chacun hydroxy, méthoxy, etc.; R<4> représente hydrogène, méthyle, benzyle, 3-isothio-cyanatopropyle, etc.; et (R<5>)m représente hydrogène, benzo substitué, etc. On a démontré que ces composés produisent d'excellents effets préventifs sur les troubles des nerfs crâniens. Ils sont par conséquents utiles comme médicaments pour inhiber divers troubles cérébraux ischémiques, hémorragiques ou traumatiques et divers troubles des nerfs crâniens causés par diverses dégénérescences nerveuses, pour traiter et prévenir diverses maladies cérébrales, telles que les attaques cérébrales, les maladies cérébrales traumatiques, les oedèmes cérébraux et les maladies dues à une dégénérescence des nerfs crâniens, pour améliorer l'état post-traumatique et pour prévenir la récurrence de tels troubles.
