Novel packaging cell lines for production of lentiviral vectors are disclosed. The cell lines utilize an ecdysone-inducible system for virus production, thereby avoiding the cytotoxic effect of unregulated expression of certain envelope proteins and lentiviral proteins. Various embodiments of the cell lines are disclosed, which are suitable for different purposes that range from in vitro screening assays to gene therapy in vivo.

L'invention concerne de nouvelles lignées cellulaires d'empaquetage pour la production de vecteurs lentiviraux. Ces lignées cellulaires utilisent un système inductible par l'ecdysone pour la production de virus, ce qui permet d'éviter l'effet cytotoxique de l'expression non régulée de certaines protéines d'enveloppe et protéines lentivirales. L'invention concerne différents modes de réalisation de ces lignées cellulaires, appropriés à différentes fins, qui vont des dosages de criblage in vitro à la thérapie génique in vivo.
