The present invention provides a clinically-applicable approach for inducing long-term, donor-specific tolerance to donor antigens, especially in recipients of CTA and/or solid organ transplants, without the requirement for patient preconditioning, without the need for chronic immunosuppressive regimens, and without the occurrence of GVHD. In particular, a method is provided for inducing donor-specific tolerance in a semi-allogeneic or a fully- allogeneic transplant recipient by administering to the recipient a therapeutically effective amount of an immunosuppressive agent that depletes T cells and a therapeutically effective amount of anti-.alpha..beta. T cell receptor antibodies, and implanting an allograft into the recipient.
L'invention porte sur une approche cliniquement applicable induisant une tolérance à long terme et spécifique du donneur aux antigènes du donneur, et spécialement chez les receveurs d'allogreffes de tissus composites (CTA) et/ou de greffons d'organes solides, et cela sans nécessiter de préconditionnement du patient, ni de régime immunosuppresseur chronique, et sans produire de réaction greffe contre hôte (GVHD). L'invention porte en particulier sur une méthode d'induction de tolérance spécifique du donneur chez un récepteur de greffons semi-allogènes ou entièrement allogènes, consistant à lui administrer une dose à efficacité thérapeutique d'un agent immunosuppresseur qui épuise les lymphocytes T, et une dose à efficacité thérapeutique d'anticorps anti-récepteur des lymphocytes T.alpha..beta., puis à procéder à l'implantation de l'allogreffe.
