The present invention provides methods for inducing insulin gene expression in cultured pancreas cells, the method comprising contacting a culture of endocrine pancreas cells expressing a PDX-1 gene with a GLP-1 receptor agonist, wherein the cells have been cultured under conditions such that the cells are in contact with other cells in the culture, thereby inducing insulin gene expression in the cells. The invention also provides high throughput screening methods for modulators of .szlig.-cell function, stable cultures of cells made by the methods of the invention, and methods of treating a human subject using the methods of the invention.

La présente invention concerne des méthodes d'induction de l'expression génétique de l'insuline dans des cellules pancréatiques cultivées, la méthode consistant à mettre en contact une culture de cellules pancréatiques d'endocrine exprimant un gène PDX-1 avec un agoniste du récepteur GLP-1, lesdites cultures ayant été cultivées dans des conditions selon lesquelles les cellules sont mises en contact avec d'autres cellules dans la culture, ce qui permet d'induire l'expression génétique de l'insuline dans les cellules. Cette invention concerne également des méthodes de criblage à rendement élevé destinées à des modulateurs de la fonction cellulaire .szlig., des cultures stables de cellules obtenues par le biais de ces méthodes, et des méthodes de traitement d'un sujet humain au moyen des méthodes de ladite invention.
