Cationic antimicrobial peptides (AMPs) are an integral part of the innate immune system. Cathelicidin and defensin homologs from a variety of species exhibit broad-range bactericidal activity. The human cathelicidin analog, hCAP18, is encoded by the CAMP gene. Vitamin D3 and its analogs upregulate transcription of CAMP and defensin ~2 (defB2) genes, leading to increased expression of hCAP18 mRNA and defB2. Induction of CAMP was observed in acute myeloid leukemia (AML), immortalized keratinocyte and colon cancer cell lines, as well as normal human bone marrow (BM)-derived macrophages and fresh BM cells. The present invention provides methods of inducing cathelicidin production by administering Vitamin D3 or Vitamin D3 analogs, as well as methods of treating skin infections and infections of the colon, sepsis and wound healing, preventing bacterial growth on skin grafts, promoting angiogenesis, and promoting chemoattraction by administering Vitamin D3 or Vitamin D3 analogs to upregulate cathelicidin and defensin expression.

Selon l'invention, les peptides cationiques antimicrobiens (PCA) font partie intégrante du système de l'immunité innée. Des homologues de cathélicidine et de défensine issus d'une variété d'espèces présentent une activité bactéricide de large portée. L'analogue de cathélicidine humaine, hCAP18, est codé par le gène CAMP. La vitamine D3 et ses analogues régulent de manière positive la transcription des gènes CAMP et défensine ß2 (defB2), ce qui augmente l'expression d'ARNm de hCAP18 et de defB2. L'induction de CAMP a été observée dans des lignées cellulaires de leucémie myéloïde aiguë (LMA), de kératinocytes immortalisés et de cancer du colon, ainsi que dans des macrophages issus d'une moelle osseuse humaine normale et dans des cellules fraîches de moelle osseuse. L'invention concerne également des procédés destinés à induire la production de cathélicidine par l'administration de vitamine D3 ou d'analogues de vitamine D3, ainsi que des méthodes de traitement des infections cutanées et des infections du colon, de la sepsis et des plaies, de prévention de la croissance bactérienne sur les greffes cutanées, de promotion de l'angiogenèse et de promotion de la chimio-attraction, par l'administration de vitamine D3 ou d'analogues de vitamine D3, de sorte à réguler de manière positive l'expression de cathélicidine et de défensine.
