The present invention is directed to the identification and use of ribonucleoside analogs to induce the mutation of an RNA virus, including HIV and HCV, or a virus which otherwise replicates through an RNA intermediate. The increase in the mutation rate of the virus results in reduced viability of progeny generations of the virus, thereby inhibiting viral replication. In addition to these methods and related compositions, the invention provides methods and combinatorial chemistry libraries for screening ribonucleoside analogs for mutagenic potential.
Cette invention se rapporte à l'identification et à l'utilisation d'analogues de ribonucléosides dans le but d'induire la mutation d'un virus d'ARN, y compris les virus VIH et VCH, ou de tout autre virus effectuant une réplication par un intermédiaire d'ARN. L'augmentation de la vitesse de mutation du virus entraîne une viabilité réduite des générations progéniques du virus, inhibant ainsi la réplication virale. En plus de ces procédés et des compositions apparentées, cette invention présente des procédés et des bibliothèques de chimie combinatoire permettant le criblage d'analogues de ribonucléosides en vue de déterminer leur potentiel mutagène.
