This invention provides a system for inhibiting the binding between two cells, one expressing P- or L-selectin on the surface and the other expressing the corresponding ligand. A covalently cross-linked lipid composition is prepared having saccharides and acidic group on separate lipids. The composition is then interposed between the cells so as to inhibit binding. Inhibition can be achieved at an effective oligosaccharide concentration as low as 10 6 fold below that of the free saccharide. Since selectins are involved in recruiting cells to sites of injury, this system can be used to palliate certain inflammatory and immunological conditions.
Cette invention concerne un système d'inhibition de la fixation entre deux cellules, l'une exprimant la sélectine P ou L sur la surface et l'autre exprimant le ligand correspondant. On prépare une composition lipidique, réticulée de manière covalente, qui renferme des saccharides et un groupe acide sur des lipides séparés. Ensuite, on interpose la composition entre les cellules pour inhiber la fixation. L'inhibition peut être obtenue à une concentration en oligosaccharides efficace, 10<6> fois inférieure au saccharide libre. Du fait que les sélectines sont impliquées dans le recrutement des cellules sur des sites de lésions, ce système peut être utilisé pour pallier certains états inflammatoires et immunologiques.
