A Protein isolated from crude extracts of Hirudo medicinalis is disclosed, which strongly inhibits the binding to collagen of platelets and their subsequent activation, which leads to platelet aggregation and thrombus formation. Additionally the protein prevents binding of von Willebrand factor Lo collagen. Described is a method for isolation and purification of the protein as well as its use for blocking collagen-stimulated platelet aggregation. The new protein (Brandinin) has a mol weight of approximately 15 kD, binds to collagen but has no collagen-cleaving activity. The protein is useful in the prophylaxis, prevention and treatment of thrombotic diseases and for coating of blood-contacting materials, rendering them thromboresistant.
L'invention se rapporte à une protéine isolée à partir d'extraits bruts d'Hirudo medicinalis qui inhibe fortement la liaison du collagène des plaquettes et l'activation ultérieure de celles-ci, laquelle conduit à l'agrégation plaquettaire et à la formation de thrombus. En outre, cette protéine empêche la liaison du facteur von Willebrand au collagène. L'invention concerne également un procédé d'isolation et de purification de la protéine en même temps que son utilisation pour bloquer l'agrégation plaquettaire stimulée par le collagène. La nouvelle protéine (Brandinin) a une masse moléculaire d'à peu près 15 kD, se lie au collagène mais n'a pas d'activité de clivage de celui-ci. Cette protéine est utile dans la prophylaxie, la prévention et le traitement des maladies thrombotiques. Cette protéine peut également être utilisée pour enrober des matériaux destinés à entrer en contact avec le sang, ladite protéine empêchant la formation d'un thrombus sur lesdits matériaux.
