The present invention relates to a method for identifying an inhibitor of a biotin protein ligase. The method include the steps of (a) providing a substrate, wherein the substrate may be biotinylated; (b) contacting the substrate with biotin and a biotin protein ligase in the presence of a test compound; (c) determining the extent of biotinylation of the substrate by the biotin protein ligase in the presence of the test compound; and (d) identifying the test compound as an inhibitor of the biotin protein ligase by a reduction in the biotinylation of the substrate in the presence of the test compound as compared to the extent of biotinylation of the substrate in the absence of the test compound.
L'invention concerne une méthode pour identifier un inhibiteur de ligase de protéine de biotine. Cette méthode comprend les étapes consistant à: (a) fournir un substrat, ce substrat pouvant être biotinylé; (b) mettre en contact le substrat avec de la biotine et avec une ligase de protéine de biotine, en présence d'un composé test; (c) déterminer l'étendue de la biotinilysation du substrat par la ligase de protéine de biotine, en présence du composé test; et (d) identifier le composé test comme inhibiteur de la ligase de protéine de biotine, par un réduction de la biotinilysation du substrat en présence du composé test, comparé à l'étendue de la biotinilysation du substrat en l'absence de ce composé test.
