The present invention pertains to novel inhibitors of cyclin-dependent kinases (CDKs), particularly CDK/cyclin complexes, which inhibitors can be used to control proliferation and/or differentiation of cells in which the inhibitors are introduced. More specifically, the inhibitors of the invention are chimeric proteins which include CDK-binding motifs from two or more different proteins. For example, the subject chimeric proteins can be generated from the in-frame fusion of coding sequences from two different CDK inhibitor proteins, such as may be derived from fusion of coding sequences for an INK4 protein and coding sequences for a CIP protein. Chimeric proteins of the present invention have been observed to be more potent inhibitors of cyclin/CDK complexes than were either of the portions of the chimeric protein individually.

La présente invention se rapporte à de nouveaux inhibiteurs des kinases dépendantes de la cycline (CDK), notamment à des complexes CDK/cycline, ces inhibiteurs pouvant être utilisés pour réguler la prolifération et/ou la différentiation des cellules dans lesquelles ils sont introduits. Plus spécifiquement, ces inhibiteurs sont des protéines chimères qui comprennent des motifs de liaison à CDK provenant d'au moins deux protéines différentes. Par exemple, ces protéines chimères peuvent être issues de la fusion au niveau du cadre de séquences codantes provenant de deux protéines inhibitrices de CDK différentes, telles que celles pouvant provenir de la fusion des séquences codant pour une protéine INK4 et des séquences codant pour une protéine CIP. Ces protéines chimères se sont avérées être des inhibiteurs des complexes cycline/CDK plus puissants que chacune des parties de la protéine chimère prises individuellement.
