Novel compounds and pharmaceutical compositions useful as EGFR tyrosine kinase inhibitors. Methods of the invention include administration of the EGFR TK inhibitors to treat diseases characterized by enhanced expression of EGF, including cancers, particularly breast cancer. Additionally, a homology model representing the structure of EGFR kinase domain is provided, which model is useful for the rationally design and screening of compounds predicted to bind favorably to EGFR and to inhibit EGFR TK.
L'invention porte: sur de nouveaux composés et préparations pharmaceutiques servant d'inhibiteurs de la tyrosine kinase du récepteur du facteur de croissance épithélial (EGFR TK); sur un procédé d'administration des inhibiteurs de la EGFR TK pour traiter des maladie caractérisées par une expression accrue d'EGF dont des cancers et en particulier les cancers du sein; et sur un modèle homologique représentant la structure du domaine des kinases du EGFR, servant à l'élaboration rationnelle et au criblage de composés prévus pour se fixer favorablement à l'EGFR afin d'inhiber la EGFR TK.
