The present invention relates to a method to limit insect and/or mite damage in plants, by the use of mutant plant proteinase inhibitors. More specifically, it relates to the use of mutant serpins of which the reactive centre loop has been replaced by an artificial sequence. Preferably, said mutant serpins use an Arabidopsis thaliana serpin-1 backbone, or a homologue thereof. Said mutant serpins have a specificity other than the wild type serpins, and by modulating the reactive loop center, specific inhibitors against specific insect and/or mite proteases can be developed. Those mutant serpins can be used to inhibit or limit insect and/or mite damage, such as the damage caused by insect feeding.
La présente invention porte sur un procédé pour limiter un endommagement par des insectes et/ou des mites dans des plantes, par l'utilisation d'inhibiteurs de protéinase de végétaux mutants. Plus spécifiquement, elle porte sur l'utilisation de serpines mutantes dont la boucle centrale réactive a été remplacée par une séquence artificielle. De préférence, lesdites serpines mutantes utilisent un squelette de serpine-1 d'Arabidopsis thaliana ou un homologue de celui-ci. Lesdites serpines mutantes ont une spécificité autre que les serpines de type sauvage, et par modulation du centre de la boucle réactive, des inhibiteurs spécifiques contre des protéases d'insectes et/ou de mites spécifiques peuvent être mis au point. Ces serpines mutantes peuvent être utilisées pour inhiber ou limiter un endommagement par des insectes et/ou des mites, tel que l'endommagement provoqué par l'alimentation d'insectes.
