In a method of targeting integration of a transgene comprising retrovirus-like DNA into a eukaryotic genome, the genome is cleared by an endonuclease and the transgene is introduced at the site of cleavage, wherein the endonuclease is specific to a site in an abundant rDNA locus and is fused to an integrase that mediates the introduction of the transgene. The fusion protein may be new.

Dans un procédé de ciblage, l'intégration d'un transgène comprend un ADN de type rétrovirus dans un génome eucaryotique, le génome étant clarifié par une endonucléase et le transgène introduit au niveau du site de clivage. L'endonucléase est spécifique à un site dans un locus ADNr abondant et est fusionné à une intégrase qui induit l'introduction du transgène. La protéine de fusion peut être nouvelle.
