The hybrization device is comprised of an oligonucleotide probe and a solid substrate wherein the solid substrate has a support surface with a neutral or negative electrostatic field and a hybridization surface. As shown in the figure, the hybridization surface is accessible for linking the oligonucleotide probe to the solid substrate. The oligonucleotide probe is linked to the hybridization surface of the solid substrate at a distance of no more than 100 angstroms. Further, there is a method of using the hybridization device to detect single base changes in DNA or RNA target sequences.
Le dispositif d'hybridation selon l'invention est constitué d'une sonde oligonucléotidique et d'un substrat solide pourvu d'une surface de support présentant un champ électrostatique neutre ou négatif et d'une surface d'hybridation. Comme le montre la figure, la surface d'hybridation est accessible pour permettre la liaison de la sonde oligonucléotidique au substrat solide. La sonde oligonucléotidique est liée à la surface d'hybridation du substrat solide à une distance ne dépassant pas 100 angströms. L'invention concerne en outre un procédé permettant d'utiliser le dispositif d'hybridation pour détecter des changements simples de bases dans des séquences cibles d'ADN ou d'ARN.
