We have performed separation of bacterial and cancer cells from peripheral human blood in microfabricated electronic chips by dielectrophoresis. The isolated cells were examined by stining the nuclei with fluorescent dye followed by laser induced fluorescence imaging. We have also released DNA and RNA from the isolated cells electronically and detected specific marker sequences by DNA amplification followed by electronic hybridization to immobilized capture probes. Efforts towards the contruction of a "laboratory- on-a-chip" system are presented which involves the selection of DNA probes, dyes, reagents and prototyping of the fully integrated portable instrument.
Nous avons procédé à la séparation de cellules bactériennes et cancéreuses de sang humain périphérique, dans des micropuces électroniques, par diélectrophorèse. Nous avons examiné les cellules isolées par coloration des noyaux avec un colorant fluorescent, puis par imagerie par fluorescence induite au laser. Nous avons également extrait de l'ADN et de l'ARN des cellules isolées, par voie électronique, et détecté des séquences marqueurs spécifiques, par amplification d'ADN, puis par hybridation électronique avec des sondes de capture immobilisées. Les efforts visant à l'élaboration d'un système de <= laboratoire sur puce >= sont présentés, notamment la sélection de sondes d'ADN, de colorants et de réactifs, ainsi que le prototypage de l'instrument totalement intégré portatif.
