The present invention provides a composition of influenza virosomes comprising reconstituted envelopes of said virus, wherein the viral envelopes are entirely derived from influenza viral particles, wherein no lipid is added from an external source to the reconstituted virosomes, wherein the virosomes comprise the influenza antigens haemagglutinin and/or neuraminidase or derivatives thereof, wherein no separate adjuvant and/or immune stimulator is added to the composition, and wherein the composition is for intranasal or inhalational administration, which composition is characterized in that a single intranasal or inhalational administration to a human being is sufficient for the induction of a systemic immune response and/or a local immune response and/or a cytotoxic lymphocytes response against said influenza antigens, which systemic response is in accordance with the CHMP criteria for influenza vaccine, and wherein the dose of haemagglutinin per viral strain per intranasal or inhalational administration is lower than or equal to 30 µg. The invention also provides the use of reconstituted influenza virosomes for the manufacture of said composition, and accordingly manufactured vaccine formulations.
La présente invention concerne une composition de virosomes de la grippe contenant des enveloppes reconstituées dudit virus. Les enveloppes virales sont entièrement dérivées de particules virales de la grippe. Aucun lipide provenant d'une source externe n'est ajouté aux virosomes reconstitués. Les virosomes renferment les antigènes de la grippe de type hémagglutinine et/ou de type neuraminidase, ou leurs dérivés. Aucun additif particulier et/ou stimulant immun n'est ajouté à la composition et celle-ci est destinée à une administration par voie intranasale ou par inhalation. La composition est caractérisée en ce qu'une seule administration à un être humain, par voie intranasale ou par inhalation, est suffisante pour induire une réaction immune systémique et/ou une réaction immune locale et/ou une réaction lymphocytaire cytotoxique contre lesdits antigènes de la grippe, la réaction systémique étant conforme aux critères du CHMP (comité des médicaments à usage humain)relatifs au vaccin contre la grippe. La dose d'hémagglutinine par souche virale destinée à l'administration par voie intranasale ou par inhalation est inférieure ou égale à 30 µg. L'invention concerne également l'utilisation de virosomes de la grippe reconstitués pour la fabrication de ladite composition, et par conséquent des formulations du vaccin.
