This invention relates to a DNA construct, methods of selecting for high- expressing host cells, a method of producing a protein of interest in high yields and a method of producing eukaryotic cells having multiple copies of a sequence encoding a protein of interest. In one method, stable clones capable of producing a high level of a product of interest are generated from one step of a direct selection immediately after transfection.

L'invention porte sur une chimère d'ADN, sur des procédés de sélection de cellules hôtes à forte expression, sur un procédé de production à fort rendement d'une protéine d'intérêt, et sur un procédé de production de cellules eucariotes présentant de multiples copies d'une séquence codant pour une protéine d'intérêt. Dans l'un des procédés, on crée des clones stables capable de produire à un niveau élevé un produit d'intérêt à partir de l'une des étapes d'une sélection directe suivant immédiatement la transfection.
