Pharmaceutical compositions comprising an invariant chain protein, or peptide fragment thereof, and a pharmaceutically acceptable carrier are disclosed. An invariant chain peptide fragment of the composition comprises an amino acid sequence of a binding motif for a major histocompatibility complex (MHC) molecule, such as an MHC class II molecule. Preferably the peptide fragment is about 10 to 35 amino acids in length, more preferably 10 to 20 amino acids in length, even more preferably 12 to 16 amino acids in length. The invention further provides a method for preparing an invariant chain peptide fragment for use in the disclosed compositions and methods. This method involves identifying an amino acid sequence of a binding motif for an MHC molecule, selecting a region of an invariant chain protein comprising the amino acid sequence of the MHC binding motif and preparing an invariant chain peptide fragment encompassing the selected region. Methods for treating autoimmune diseases are also disclosed. The methods of the invention involve administering to a subject in need of treatment a therapeutically effective amount of an invariant chain protein, or peptide fragment thereof, optionally with a pharmaceutically acceptable carrier.

Un fragment peptidique à chaîne invariable de la composition comprend une séquence d'acides aminés d'un motif de liaison pour une molécule du complexe majeur d'histocompatibilité (HL-A), telle qu'une molécule de classe II du HL-A. De préférence, le fragment peptidique comporte en longueur environ 10 à 35 acides aminés, de préférence 10 à 20 acides aminés, et plus avantageusement 12 à 16 acides aminés. L'invention se rapporte également à un procédé de préparation d'un fragment peptidique à chaîne invariable utilisé dans les compositions et procédés décrits. Ce procédé consiste à identifier une séquence d'acides aminés d'un motif de liaison pour une molécule de HL-A, à sélectionner une région d'un protéine à chaîne invariable comprenant la séquence d'acides aminés du motif de liaison de HL-A et à préparer un fragment peptidique à chaîne invariable renfermant la région sélectionnée. Les procédés de l'invention consistent à administrer à un sujet nécessitant ce traitement une quantité thérapeutiquement efficace d'une protéine à chaîne invariable, ou un fragment peptidique de celle-ci, éventuellement avec un excipient pharmaceutiquement acceptable.
