The present invention is directed to a bacterial delivery system for delivering alphavirus replicon DNA into an animal or animal cells with the replicon encoding one or more heterologous genes to be expressed in the animal or the animal cells. The bacteria are invasive bacteria or attenuated invasive bacteria engineered to contain a DNA vector that encodes the alphavirus replicon in an eukaryotic expression cassette. The heterologous gene can encode an antigen, a therapeutic agent, an immunoregulatory agent, an anti- sense RNA, a catalytic RNA, a protein, a peptide, an antibody or an antigen- binding fragment of an antibody. In a preferred embodiment, the heterologous gene encodes one or more antigens useful as a vaccine for HIV. In addition to the bacterial delivery system, the invention provides methods of introducing and expressing the heterologous gene(s) in animal or animal cells and methods of stimulating or inducing an immune response.
L'invention concerne un système d'administration bactérien destiné à administrer un ADN de réplicon d'alphavirus à un animal ou dans des cellules animales, ledit réplicon codant un ou plusieurs gènes hétérologues exprimés chez l'animal ou dans les cellules animales. Les bactéries se présentent comme des bactéries invasives ou des bactéries invasives atténuées auxquelles on a appliqué des opérations de génie génétique pour qu'elles contiennent un vecteur d'ADN codant le réplicon d'alphavirus dans une cassette d'expression eucaryote. Le gène hétérologue peut coder un antigène, un agent thérapeutique, un agent immunorégulateur, un ARN antisens, un ARN catalytique, une protéine, un peptide, un anticorps ou un fragment de liaison d'antigènes d'un anticorps. Dans un mode de réalisation préféré, le gène hétérologue code un ou plusieurs antigènes utiles en tant que vaccin contre le VIH. Outre le système d'administration bactérien, le système concerne des procédés d'introduction et d'expression d'un ou de plusieurs gènes hétérologues chez un animal ou dans des cellules animales et des procédés pour stimuler ou induire une réponse immunitaire.
