A process for disinfecting protein solutions operates by flowing the solutions through a bed of iodinated organic polymer that acts as a gel filtration material. The preferred types of gel filtration material is Sephadex brand cross-linked dextrans, either G-25 or A-25 (DEAE modified Sephadex). The various types of cross-linked dextrans are readily iodinated before use by being mixed dry with elemental iodine at an elevated temperature such as 37 ~C. Sephadex brand cross-linked dextrans can be readily iodinated to 50 % weight of iodine to weight of dextrans or more. The preferred level of iodination is 30-40 %. Before use the sephadex is first swelled in aqueous medium and then poured into a chromatographic column. Six ml of swelled 30 % iodine Sephadex A-25 can effectively destroy virus in at least one liter of 5 % human IgG.
L'invention porte sur un procédé de désinfection des solutions protéinées consistant à faire passer les solutions dans un lit de polymère organique iodé faisant office de matière de filtration de gel. Les types de matière de filtration de gel utilisés de préférence sont les dextranes réticulés de la marque Sephadex, soit le type G-25, soit le type A-25 (Sephadex modifié de type diéthyl-aminoéthyl - DEAE). Les divers types de dextranes réticulés sont aisément iodés avant utilisation par un procédé de mélange à sec de ces dextranes avec de l'iode élémentaire, à une température élevée, par exemple à 37·. L'iodation des dextranes réticulés de la marque Sephadex se fait aisément par l'apport d'iode de 50 % en poids, voire plus, par rapport au poids des dextranes. Le niveau d'iodation préféré est compris entre 30 et 40 %. Avant utilisation, le Sephadex est augmenté d'iode dans un milieu aqueux, puis déversé dans une colonne chromatographique. Six ml de Sephadex de type A-25 augmenté de 30 % d'iode peuvent détruire efficacement un virus contenu dans au moins un litre de solution d'IgG humaine à 5 %.
