The subject invention provides an isolated protein having an apparent molecular weight of about 27 kD and capable of binding to and inhibiting the activation of a cyclin E-Cdk2 complex. The subject invention also provides the nucleic acid encoding the p27 protein, vectors, and host cells for producing the recombinant form of the p27 protein. The subject invention further provides methods for identifying agents capable of modulating the ability of p27 to inhibit the activation of the cyclin E-Cdk2 complex. Finally, the subject invention provides methods for treating subjects diagnosed with hypoproliferative disorders using said agents.
Cette invention se rapporte à une protéine isolée qui possède un poids moléculaire apparent d'environ 27 kD et qui est capable de se lier à un complexe cycline E/Cdk2 et d'inhiber l'activation d'un tel complexe. Cette invention se rapporte également à l'acide nucléique codant cette protéine p27, à des vecteurs et à des cellules hôtes servant à produire la forme recombinée de cette protéine p27. Cette invention se rapporte en outre à des procédés pour identifier des agents capables de moduler la capacité de la protéine p27 à inhiber l'activation du complexe cycline E/Cdk2. Cette invention se rapporte enfin à des procédés utilisant de tels agents pour traiter les sujets sur lesquels on a diagnostiqué des troubles hypoprolifératifs.
