The method of the invention is directed to the novel use of a diffusion chamber within which previously "uncultivatible" microorganisms can be isolated. Rather than attempting to replicate the natural environment of an unknown microorganism, the method of the invention provides for exposing dividing microorganisms to all the components of the original environment while simultaneously containing the resulting colonies so that they can be isolated. The method of the invention can take advantage of the recognition that the preponderance of difficult-to-grow microorganisms do not form colonies visible to the naked eye. Therefore, these organisms must be isolated under a compound microscope as "microcolonies". In addition, methods according to the invention permit the isolation of novel microorganisms capable of growing in artificial media only in co-culture in the presence of a companion microorganism.
La présente invention concerne une nouvelle utilisation d'une chambre de diffusion à l'intérieur de laquelle peuvent être isolés des microorganismes qui étaient jusque là <= non cultivables >=. Plutôt que de tenter de reproduire l'environnement naturel d'un microorganisme inconnu, le procédé de l'invention choisit d'exposer des microorganismes en division à tous les composants de leur environnement d'origine tout en contenant simultanément les colonies obtenues afin de pouvoir les isoler. Le procédé précité tire parti du fait que la prédominance de microorganismes difficiles à cultiver ne forme pas de colonies visibles à l'oeil nu. C'est pourquoi ces organismes doivent être isolés sous un microscope composé, sous la forme de <= microcolonies >=. L'invention se rapporteen outre à des procédés qui permettent d'isoler de nouveaux microorganismes capables de croître dans un milieu artificiel uniquement en co-culture avec un microorganisme compagnon.
