A method is disclosed permitting the determination of comprehensive immunoglobulin-specific reaction profiles of people with immuno-functional disorders by means of the individual V gene repertory of Ig-expressing B- cells. The method comprises the isolation of B-cells from body fluids, the isolation of individual B-cells and the genetic material thereof, the amplification of nucleic acids, coding for the variable regions of an antibody expressed by a B-cell, the recombinant production of antibodies by expression of the amplified material and the determination of the binding of the recombinant antibody to antigens.
La présente invention concerne un procédé qui permet l'établissement de profils réactionnels spécifiques de l'immunoglobuline, globaux, de personnes soufrant de troubles immunitaires, grâce au répertoire de gène V individuel de lymphocytes B exprimant Ig. Le procédé comprend l'isolation de lymphocytes B dans des fluides corporels, l'isolation de lymphocytes B individuels et de leur matériel génétique, l'amplification d'acides nucléiques qui codent pour les domaines variables d'un anticorps exprimé par un lymphocyte B, la production recombinée d'anticorps grâce à l'expression des acides nucléiques amplifiés, et la détermination de la capacité de liaison de l'anticorps recombiné, pour des antigènes.
