The invention relates to a method for the stabilisation, purification or/and isolation of nucleic acids from material samples, in particular, stool samples, which can contain impurities and inhibitors or interfering substances. The invention further relates to a reagent kit for carrying out said method. The basis of the invention is, in particular, a method for purification, stabilisation or/and isolation of nucleic acids from material samples, whereby a buffer is added to the sample containing the nucleic acids, with a pH value of 2 to 7, a salt concentration of at least 100mM, or/and a phenol neutralising substance. According to the invention, pure nucleic acids which may be amplified can be obtained from faecal samples by a simple method, which are suitable for diagnostic proof of infections, in particular, bacterial or viral infections, or mutations, in particular, for tumour-specific DNA mutations.
L'invention concerne un procédé de stabilisation, purification et/ou d'isolement d'acides nucléiques contenus dans des échantillons de matières, en particulier des échantillons de selles pouvant contenir des impuretés et des inhibiteurs ou des substances parasites. L'invention concerne également une trousse de réactifs à utiliser pour la mise en oeuvre de ce procédé. L'objectif de l'invention est en particulier un procédé de purification, de stabilisation et/ou d'isolation d'acides nucléiques contenus dans des échantillons de matières, selon lequel on ajoute à l'échantillon contenant les acides nucléiques un tampon qui présente un pH de 2 à 7 et une concentration de sel d'au moins 100 mM et/ou contient une substance neutralisant le phénol. Le procédé selon l'invention permet, de façon simplifiée, d'obtenir, à partir d'échantillons de matières fécales, des acides nucléiques purs et pouvant être amplifiés, lesquels peuvent être utilisés pour le diagnostic d'infections, en particulier d'infections bactériennes ou virales, ou de mutations, en particulier de mutations d'ADN spécifiques des tumeurs.
