The present invention relates to materials for extracting nucleic acid using charge switch methods which involve, at a first pH, bringing the sample into contact with a material which comprises an ionisable group, wherein the material has a positive charge at said first pH, such that nucleic acid is bound to the material; and releasing the nucleic acid at a second, higher, pH at which the charge on the material is negative, neutral or less positive. The release of the nucleic acid occurs under mild conditions and the ionisable group is provided by a biological buffer, a polyhydroxylated amine, a detergent or surfactant, a carbohydrate, a nucleic acid base, a heterocyclic nitrogen-containing compound, a monoamine, a biological dye, or a negatively ionisable group, the pKa of which is between about 3.0 and 7.0 and a metal oxide which is positively charged at said first pH, and optionally also at said second pH.
L'invention concerne des matériaux destinés à extraire un acide nucléique au moyen de méthodes de changement de charge consistant, à un premier pH, à amener l'échantillon en contact avec un matériau comprenant un groupe ionisable, ce matériau possédant à ce premier pH une charge positive, de telle façon que l'acide nucléique se lie au matériau, puis à relarguer l'acide nucléique à un second pH, plus élevé, pH auquel la charge sur le matériau devient négative, neutre ou moins positive. Le relargage de l'acide nucléique se produit dans des conditions douces et le groupe ionisable est porté par un tampon biologique, une amine polyhydroxylée, un détergent ou un tensioactif, un carbohydrate, une base d'acide nucléique, un composé à hétérocycle azoté, une monoamine, un colorant biologique ou un groupe à ionisation négative, dont la pKa est compris entre 3,0 et 7,0 environ, et un oxyde métallique, positivement chargé au premier pH, et éventuellement aussi au second pH.
