Methods for amplifying target nucleic acid sequences using a nucleic acid polymerase lacking 5' exonuclease activity and a set of oligonucleotide primers. Preferably, a primer array is used. The primer array contains two sets of primers. One set contains at least two complementary primers, The other set contains at least two sense primers. Using the described methods, amplification can be carried out under essentialy constant environmental conditions without the requirement for exonuclease activity or restriction endonuclease activity.

Méthodes d'amplification de séquences d'acide nucléique cibles au moyen d'une polymérase d'acide nucléique dénuée d'activité endonucléase 5', et d'un ensemble d'amorces oligonucléotidiques. On utilise, de préférence, un groupe d'amorces. Un ensemble contient au moins deux amorces complémentaires, alors que l'autre contient au moins deux amorces sens. Grâce à ces méthodes, il est possible de procéder à l'amplification dans des conditions environmentales pratiquement constantes sans que l'activité d'exonucléase ou de d'endonucléase de restriction ne soit nécessaire.
