Isoxazolecarboxamide derivatives (I) (R=alkyl, alkoxy, polyfluoroalkoxy, OH or CF3SO2O; each of R1 and R2 independently =H, halogen, polyfluoroalkoxy or alkoxy; R3 = one or more substituents selected from H, halogen, alkyl, alkoxy, NO2, NH2, NHacyl, CN, alkoxycarbonyl, carboxamido; R4 = H, alkyl or aralkyl; an n is 0, 1 or 2) and their N-oxides and pharmaceutically acceptable salts are endowed with adrenergic antagonist activity and high selectivity toward the .alpha.la adrenergic receptor with respect to the 5-HTlA receptor. This activity profile suggests the use of these derivatives in the treatment of obstructive syndromes of the lower urinary tract, including BPH, without side effects associated with hypotensive activity being present.

L'invention concerne des dérivés d'isoxazolecarboxamide représentés par la formule générale (I) dans laquelle R représente alkyle, alcoxy, polyfluoroalcoxy, OH ou CF¿3?SO¿2?O, chacun des R¿1? et R¿2? représente indépendamment H, halogène, polyfluoroalcoxy ou alcoxy ; R¿3? représente un ou plusieurs substituants sélectionnés parmi H, halogène, alkyle, alcoxy, NO¿2?, NH¿2?, NHacyle, CN, alcoxycarbonyle, carboxamido; R¿4? représente H, alkyle ou aralkyle; et n vaut 0, 1 ou 2), ainsi que leurs N-oxydes et sels pharmaceutiquement acceptables desdits dérivés. Ces substances présentent une activité antagoniste adrénergique et une forte sélectivité pour le récepteur .alpha.¿1a? adrénergique par rapport au récepteur 5-HT¿1A?. Ce profil d'activité suggère l'utilisation de ces dérivés dans le traitement de syndromes obstructifs des voies urinaires inférieures, y compris l'adénome prostatique, sans qu'il y ait présence d'effets secondaires associés à une activité hypotensive.
