Disclosed is a kinase and phosphatase assay based on Fluorescence Resonance Energy Transfer (FRET). The assay involves; dephosphorylating a phosphorylated substrate molecule labelled with an energy acceptor of a FRET pair with a phosphatase; contacting the dephosphorylated substate molecule with an energy donor of the FRET pair and a binding partner that only binds phosphorylated substrate molecule and the detection molecule. The contacting of the substrate molecule with the detection molecule and the binding partner result in binding of the phosphorylated substrate molecule and the detection molecule to the binding partner. The assay also includes a step of determining phosphatase activity by measuring energy transfer from the energy donor of the phosphorylated substrate molecule bound to the binding partner to the energy acceptor of the detection molecule bound to the binding partners. The assay can be useful for screening for compounds that can modulate specific signalling pathways.
On divulgue un essai de kinase et de phosphatase fondé sur le transfert d'énergie de résonance de fluorescence (TERF). L'essai comporte la déphosphorylation d'une molécule substrat phosphorylée marquée avec un accepteur d'énergie d'une paire de TERF avec une phosphatase; la mise en contact de la molécule substrat déphosphorylée avec un donneur d'énergie d'une paire de TERF et un partenaire de liaison qui ne se lie qu'à la molécule substrat phosphorylée et à la molécule de détection.. La mise en contact de la molécule substrat avec la molécule de détection et le partenaire de liaison entraîne la liaison de la molécule substrat phosphorylée et de la molécule de détection avec le partenaire de liaison. L'essai comprend aussi une étape de détermination de l'activité de la phosphatase par la mesure du transfert d'énergie du donneur d'énergie de la molécule substrat phosphorylée liée au partenaire de liaison à l'accepteur d'énergie de la molécule de détection liée au partenaire de liaison. L'essai peut être utile pour dépister des composés capables de moduler certains mécanismes de signalisation.
