One aspect of the present invention relates to a method for the kinetic resolution of racemic and diastereomeric mixtures of chiral compounds. The critical elements of the method are: a non-racemic chiral tertiary-amine- containing catalyst; a racemic or diastereomeric mixture of a chiral substrate, e.g., a cyclic carbonate or cyclic carbamate; and a nucleophile, e.g., an alcohol, amine or thiol. A preferred embodiment of the present invention relates to a method for achieving the kinetic resolution of racemic and diastereomeric mixtures of derivatives of .alpha.- and .beta.-amino, hydroxy, and thio carboxylic acids. In certain embodiments, the methods of the present invention achieve dynamic kinetic resolution of a racemic or diastereomeric mixture of a substrate, i.e., a kinetic resolution wherein the yield of the resolved enantiomer or diastereomer, respectively, exceeds the amount present in the original mixture due to the in situ equilibration of the enantiomers or diastereomers under the reaction conditions prior to the resolution step.
La présente invention concerne sous l'un de ses aspects un procédé de résolution cinétique de mélanges racémiques et diastéréomères de composés chiraux. Les agents clés du procédé sont d'abord un catalyseur chiral non-racémique contenant une amine tertiaire, ensuite un mélange diastéréomère ou racémique à base d'un substrat chiral tel qu'un carbonate cyclique ou un carbamate cyclique, et enfin un nucléophile tel qu'un alcool, une amine ou un thiol. L'un des modes préférés de réalisation de l'invention porte sur un procédé destiné à la réalisation de la résolution cinétique de mélanges racémiques et diastéréomères de dérivés d'acides carboxyliques de .alpha.- et .beta.-amino, d'hydroxy, et de thio. Pour certains modes de réalisation, les procédés de la présente invention donne une résolution cinétique dynamique d'un mélange racémique ou diastéréomère à base d'un substrat, c'est-à-dire une résolution cinétique dans laquelle le rendement de l'énantiomère ou du diastéréomère résolu, suivant le cas, dépasse la quantité présente dans le mélange original en raison de l'équilibrage in situ des énantiomères ou diastéréomères dans les conditions de réaction avant l'étape de résolution.
