The present invention relates to a kit for the labelling of biomolecules bearing reactive amino or hydroxyl groups. The kit consists of a Reagent A and a Reagent B, individually packaged, comprising: a mixture of a carboxylated labelling compound and a tertiary amine (Reagent A); and a coupling reagent (Reagent B). Upon contacting Reagent A with Reagent B, the carboxylated labelling compound is activated in-situ by the coupling reagent (a guanidinium, or uronium salt) in the presence of the tertiary amine. The active form of the carboxylated labelling compound is reacted with a biomolecules bearing reactive amino or hydroxyl groups, with formation of a stable covalent bond between the carboxylated labelling compound and the biomolecule.
