ABSTRACT A method for determining the amount of analyte in a sample. The method comprises providing a first assay compartment and a second assay compartment. The compartments are separated by a barrier permeable to a signal providing binding reagent-label conjugate. An analyte containing sample is combined with a binding reagent-label conjugate capable of binding to the analyte to form alone or, in combination with a second binding reagent, a specific binding pair complex of a size sufficiently large to prevent passage through the barrier. The binding reagent-label conjugate is provided in an excess of the amount required to bind with all the analyte. The mixture is incubated to allow the analyte to bind to the binding reagent-label conjugate to form the specific binding pair comprising the analyte and the binding reagent-label conjugate and for unbound binding reagent-label conjugate to pass through the barrier into the second compartment. The unbound binding reagent- label conjugate is entrapped and modified in the second compartment to enable detection of a signal from the specific binding pair as a measure of the amount of analyte bound in the specific binding pair. The detectable signal is measured.
