This invention relates to the area of systemic, tissue-specific non-viral gene delivery. The present invention provides a novel method to prepare ligand- directed, PEG-stabilized complex as gene delivery systems for targeted gene therapy. Due to the presence of PEG these novel complexes have longer circulating times than conventional ligand-liposome complexes. In addition, due to the presence of the ligand in the complex with PEG, these complexes are tissue targeting. Their small size further makes them very desirable for in vivo use.
Cette invention porte sur la zone d'administration systémique de gènes non viraux spécifiques d'un tissu. Cette invention porte également sur un nouveau procédé de préparation d'un complexe stabilisé par PEG, dirigé sur un ligand selon des systèmes d'administration de gènes utilisés en thérapie génique ciblée. Du fait de la présence de PEG, ces nouveaux complexes ont des temps de circulation plus longs que les complexes ligand-liposome traditionnels. De plus, du fait de la présence du ligand dans le complexe avec le PEG, ces complexes sont ciblés sur les tissus. Grâce à leur petite taille, il est possible de les utiliser in vivo.
