The invention comprises a light profile microscopy (LPM) apparatus and method used for the direct three dimensional micron scale imaging of single biological cells or cellular arrays. A slice volume is illuminated in a cellular sample using a planar sheet of optical radiation of micron scale thickness. A microscope records a full field image of the lateral dimensions of the cellular slice volume with optical contrast from mechanisms that emit or scatter light from the cellular slice volume, including luminescence, Raman scattering and multi- photon processes. Imaging occurs with effective refractive index matching to an immersion objective, and image focus is independently confirmed by a registration beam system. A series of axially distributed slice volume images is recorded, de-convoluted and assembled into a tomogram of the cellular object. This method provides two orders of magnitude shorter image acquisition times over the prior art with a lower photon dosage per image element.
