An immunoassay for detecting lipopolysaccharides from Gram negative bacteria such as E.coli, Chlamydia or Salmonella and a device for conducting the same wherein the assay comprises the use of a lipopolysaccharide binding protein and an antibody having specific binding affinity to the lipopolysaccharide analyte as first or second binding reagents. The analyte is reacted with a first binding reagent to form a complex and then reacted with the second binding reagent to form a complex comprising the analyte and first and second binding reagents. The presence of the final complex is then detected. One of the binding reagents is labelled and free, the other binding reagent is unlabelled and immobilised on a solid support. The device comprises a first zone from where the free labelled first binding reagent is carried with the sample to the detection zone comprising the unlabelled second binding reagent via capillary action.

L'invention concerne un immunoessai permettant de détecter des lipopolysaccharides à partir de bactéries Gram négatif comme E. coli, Chlamydia, ou Salmonella. Cette invention concerne également un dispositif destiné à effectuer cet immunoessai, ce dispositif s'appuyant notamment sur l'utilisation d'une protéine de liaison lipopolysaccharidique et d'un anticorps présentant une affinité de liaison spécifique à un analyte lipopolysaccharidique, cette protéine et cet anticorps servant de premier et second réactifs de liaison. On met cet analyte en présence d'un premier réactif de liaison de manière à former un complexe, cet analyte étant ensuite mis en présence du second réactif de liaison afin de former un complexe qui renferme l'analyte et le premier et le second réactifs de liaison. On peut ensuite détecter la présence du complexe ainsi obtenu. De plus, l'un desdits réactifs de liaison est marqué et libre, l'autre réactif de liaison étant non marqué et immobilisé sur un support solide. Enfin, le dispositif de cette invention comporte une première zone à partir de laquelle le premier réactif de liaison marqué et libre est amené avec l'échantillon, par action capillaire, vers la zone de détection renfermant le second réactif de liaison non marqué.
