Novel glucocorticoid receptor ligands as provided which have general formula (I) in which: R is a hydrogen atom, an aliphatic hydrocarbon, an aromatic hydrocarbon, carboxylic acid or ester thereof, alkenyl carboxylic acid or ester thereof, hydroxy, halogen, or cyano halogen, or cyano, or a pharmaceutically acceptable salt thereof. W is a methine carbon atom (CH) having the R, S, or racemic stereochemistry. X and Z are the same or are different and are a bond, an amide (-CONR'- or -NR'CO-), an amine (-NR'-), and ether (-O-), or a thioether (-S-) and R' is a hydrogen atom, an aliphatic hydrocarbon, or an aromatic hydrocarbon. The subscripts n, o are the same or are different and are integers from one (1) to six (6). m is an integer from zero to 6. Y is hydroxyl group, carboxylic acid or ester thereof, a tetrazole, an acylsulfonamide (-CONHSO2R" or -SO2NHCOR" where R" is an aliphatic or aromatic hydrocarbon), or a pharmaceutically acceptable salt thereof. A method for treating diseases associated with metabolism dysfunction or which are dependent on the expression of a glucocorticoid such as diabetes.
L'invention porte sur de nouveaux ligands des récepteurs de glucocorticoïdes ayant la formule générale (I) dans laquelle R est un atome d'hydrogène, un hydrocarbure aliphatique, un hydrocarbure aromatique, un acide carboxylique ou un ester dudit acide, un acide carboxylique alcoydénique ou un ester dudit acide, un hydroxyle, un halogène, ou un cyanohalogène, ou cyano, ou un sel pharmaceutiquement acceptable desdits composés. W est un atome de carbone de méthine (CH) ayant R, S, ou une stéréochimie racémique. X et Z sont identiques ou différents et forment une liaison, un amide (-CONR'- ou -NR'CO-), une amine (-NR'-), et un éther (-O-), ou un thioéther (-S-) et R' est un atome d'hydrogène, un hydrocarbure aliphatique ou un hydrocarbure aromatique. Les indices n et o sont identiques ou sont différents et sont des entiers de un (1) à six (6). m est un entier de zéro à 6. Y est un groupe hydroxyle, un acide carboxylique ou un ester dudit acide, un tétrazole, un acyle sulfonamide (-SONHSO2R" ou SO2NHCOR" où R" est un hydrocarbure aliphatique ou aromatique), ou un sel pharmaceutiquement acceptable desdits composés. L'invention porte également sur une méthode pour le traitement de maladies liées au dysfonctionnement du métabolisme ou dépendant de l'expression d'un glucocorticoïde, telles que le diabète.
