Growth factors and/or angiogenic factors are administered in combination with dissociated cells to be transplanted, preferably in microspheres with the cells on or in a polymeric matrix, to enhance survival and profileration of the transplanted cells. Examples demonstrate that epidermal growth factor (EGF) was incorporated into microspheres fabricated from a copolymer of lactic and glycolic acid using a double emulsion technique, the incorporated EGF was steadily released over one month in vitro, and it remained biologically active, as determined by its ability to stimulate DNA synthesis, division, and long-term survival of cultured hepatocytes. EGF-containing microspheres were mixed with a suspension of hepatocytes, seeded onto porous sponges, and implanted into the mesentery of two groups of Lewis rats, to demonstrate efficacy in vivo. Two weeks after implantation in PCS animals, devices which included EGF-containing microspheres showed a two-fold increase in the number of engrafted hepatocytes, as compared to implants which received blank microspheres.
Des facteurs de croissance et/ou angiogéniques sont administrés en combinaison avec des cellules dissociées à transplanter, placées de préférence dans des microsphères, sur ou dans une matrice polymère, pour favoriser la survie et la prolifération des cellules transplantées. Des exemples portent sur un facteur de croissance de l'épiderme (EGF) incorporé dans des microphères fabriquées à partir d'un copolymère d'acide lactique et glycolique par une technique à double émulsion. L'EGF incorporé a été régulièrement libéré in vitro pendant un mois et il est resté biologiquement actif, comme déterminé par sa capacité à stimuler la synthèse de l'ADN, et la division et la survie à long terme des hépatocytes cultivés. Les microsphères contenant l'EGF ont été mélangées avec des hépatocytes en suspension, semées sur des éponges poreuses et implantées dans le mésentère de deux groupes de rats de Lewis pour démontrer leur efficacité in vivo. Deux semaines après implantation chez ceux des animaux ayant subi un anastomose portocave, les supports des microsphères contenant de l'EGF ont fait apparaître un doublement du nombre d'hépatocytes greffés par rapport aux implants qui avaient reçu des microsphères à blanc.
