Antisense oligonucleotide gene therapy selective for the 5' region of PDGFR-.beta. subunit mRNA was used in attempt to prevent intimal thickening following rat carotid arterial injury. Sustained perivascular application of the antisense oligomers for 14 days reduced PDGFR-.beta. protein overexpression and prevented neointima formation by 80%. Specificity was verified by the absence of effects on the expression of a non-targeted gene PDGFR-.alpha.. These data demonstrated that antisense oligonucleotide sequences can effectively suppress a growth factor receptor, and the reduction of intimal hyperplasia after injury correlates with the extent to which these oligomers inhibited PDGFR-.beta. protein expression. Methods and materials useful for preventing restenosis are described and claimed.
Thérapie génique à base d'oligonucléotide antisens sélectif pour la région 5' du récepteur bêta du facteur de croissance dérivé des plaquettes (PDGFR-bêta). De l'ARNm sous-unitaire a été utilisé pour tenter de prévenir l'épaississement de l'intima consécutive à une lésion de l'artère carotide chez le rat. L'application périvasculaire prolongée des oligomères antisens pendant une période de 14 jours a entraîné une diminution de la surexpression de la protéine PDGFR-bêta et a empêché dans une proportion de 80 % la formation de néo-intima. La spécificité a été vérifiée par l'absence d'effet sur l'expression du gène de PDFGR-alpha non ciblé par cette thérapie. Ces données ont montré que des séquences d'oligonucléotides antisens peuvent supprimer efficacement un récepteur d'un facteur de croissance et la réduction de l'hyperplasie de l'intima consécutive à une lésion correspond à la mesure dans laquelle ces oligomères ont inhibé l'expression de la protéine PDGFR-bêta. Des méthodes et des matériaux permettant de prévenir la resténose sont décrits et revendiqués.
