Prodrugs and pharmaceutically acceptable salts thereof are provided carrying functional groups sensitive to mild bases and capable of slowly hydrolyzing to the original active drug molecule under physiological conditions. The prodrugs are preferably of the formula X - Y wherein Y is a moiety of a drug bearing at least one functional group selected from free amino, carboxyl, hydroxyl and/or mercapto, and X is a radical selected from radicals of formulas (i) to (iv) wherein R1 and R2, the same or different, are each hydrogen, alkyl, alkoxy, alkoxyalkyl, aryl, alkaryl, aralkyl, halogen, nitro, sulfo, amino, ammonium, carboxyl, PO3H2, or OPO3H2; R3 and R4, the same or different, are each hydrogen, alkyl or aryl; and A is a covalent bond when the radical is linked to a carboxyl or mercapto group of the drug Y, or A is OCO- when the radical is linked to an amino or hydroxyl group of Y, and pharmaceutically acceptable salts thereof. Examples of such prodrugs are those wherein Y is a moiety of insulin, human or bovine growth hormone, antibiotics, propanolol and others.
Promédicaments et leurs sels pharmaceutiquement acceptables portant des groupes fonctionnels sensibles aux bases faibles et aptes à se transformer par hydrolyse dans des conditions physiologiques pour produire le médicament actif initial. De préférence, ces promédicaments répondent à la formule X-Y, dans laquelle Y représente une fraction d'un médicament portant au moins un groupe fonctionnel choisi parmi amino libre, carboxyle, hydroxyle et/ou mercapto; et X représente un radical choisi parmi les radicaux répondant aux formules (i) à (iv), dans lesquelles R1 et R2, identiques ou différents, représentent chacun hydrogène, alkyle, alcoxy, alcoxyalkyle, aryle, alkaryle, aralkyle, halogène, nitro, sulfo, amino, ammonium, carboxyle, PO3H2 ou OPO3H2; R3 et R4, identiques ou différents, représentent chacun hydrogène, alkyle ou aryle; et A représente une liaison covalente lorsque le radical est lié à un groupe carboxyle ou mercapto du médicament Y, ou A représente OCO- lorsque le radical est lié à un groupe amino ou hydroxyle du médicament Y, et leurs sels pharmaceutiquement acceptables. Lesdits promédicaments sont, par exemple, ceux dans lesquels Y représente, entre autres, une fraction d'insuline, l'hormone de croissance humaine ou bovine, des antibiotiques, le propanolol.
