The invention relates to the identification of the molecular basis of acquired long QT syndrome. More specifically, the invention has identified that HERG is associated with acquired long QT syndrome. HERG has been identified to encode the major subunit for the IKr channel. It has been found that increasing the extracellular levels of K+ paradoxically increases the outward current of this channel. Increasing extrarcellular K+ levels in patients, especially those being treated with antiarrhythmic medication or other medication which can cause cardiac arrhythmia or those with the hereditary form of long QT syndrome, decreases the likelihood of developing LQT and torsade de pointes.
L'invention porte sur l'identification du fondement moléculaire du syndrome du QT long acquis. Plus précisément, l'invention a établi que le gène HERG est associé au syndrome du QT long. On a démontré que HERG encode la principale sous-unité du canal 1Kr. On a découvert que le fait d'accroître les concentrations extracellulaires de K+ a pour effet paradoxal d'augmenter le courant centrifuge de ce canal. Le fait d'augmenter les concentration extracellulaires de potassium chez les patients, en particulier chez ceux qui reçoivent des antiarythmiques ou d'autres médicaments qui peuvent induire une arythmie cardiaque ou chez ceux qui sont atteints de la forme héréditaire du syndrome du QT long, a pour effet de réduire la probabilité de développer un syndrome du QT long et des torsades de pointe.
